چکیده

A simple, sensitive and specific HPLC method was validated for the simultaneous determination of benzoic acid and sorbic acid in noodles. Sample preparation involved the extraction with methanol and water (60:40, v/v). The separation was achieved on a GerminiC18 modified silica column (50 mm x 4.6 mm i.d., 5 μm particle diameter). The mobile phase consisted of 0.05 M ammonium acetate (pH 4.4) and methanol in the ratio of 60:40 (% v/v) at a flow rate of 1 mL/min and detection was performed at 234 nm using a diode-array detector (DAD). The method was validated with respect to specificity, linearity, accuracy, precision, limit of detection (LOD), limit of quantitation (LOQ) and robustness. The calibration curve for benzoic acid and sorbic acid was found to be linear in the concentration range of 5-200 μg/mL (r2 0.9998) and 1-50 μg/mL (r2 0.9998), respectively. The values of LOD and LOQ were 0.42 and 1.14 μg/mL and 0.32 and 0.99 μg/mL for benzoic acid and sorbic acid, respectively. The accuracy determined by spike recovery measurement was 85.61-102.04% for benzoic acid and 83.62-102.47% for sorbic acid. The method and intermediate precision for benzoic acid and sorbic acid were demonstrated by the relative standard deviation of 1.84% and 2.40% and 1.41% and 2.80%, respectively. The validation acceptance criteria were met in all cases. The proposed method was successively applied for monitoring of benzoic acid and sorbic acid in 5 different kinds of noodles.

Non Aqueous Titration for the Determination of Azelaic Acid: Introduction and Method Validation

Alpha-hydroxylic acids are widely used in the treatment and control of skin disorders. Azelaic acid is a naturally occuring Alpha-hydroxy acid that can be formed endogenously from long-chain dicardoxylic acids, metabolism of oleic acid or omega oxidation of mono carboxylic acids. It shows bacteriostatic and bactericidal activities against aerobic and anaerobic micro-organisms present on acne-be...

Analysis of benzoic acid and sorbic acid in Thai rice wines and distillates by solid-phase sorbent extraction and high-performance liquid chromatography

An application of solid-phase extraction (SPE) for high-performance liquid chromatography (HPLC) analysis was developed for the rapid determination of benzoic acid and sorbic acid in Thai rice wine and distillate products. The SPE extraction gave promising results with 95.7+1.7% recovery for benzoic acid standard, and 95.2+2.3% for sorbic acid standard. Mean relative standard deviation (RSD) fo...

IMPROVED CHROMATOGRAPHIC METHOD FOR DETERMINATION OF SODIUM DICLOFENAC IN INJECTABLE SOLUTION AND PREDICTION OF CHEMICAL STABILITY

Two modified methods for assaying sodium diclofenac were developed by GC and HPLC. Diclofenac was converted into methyl ester derivative by methyl iodide in acetone. The ester was extract and subjected to GLC with flame ionization detector. %5 SE-30/chrom W-HP (80-100 mesh) was used as a column in GC. For reversed phase HPLC, the mobile phase was methanol and water (55-45). The separation was p...

Simultaneous determination of preservatives (benzoic acid, sorbic acid, methylparaben and propylparaben) in foodstuffs using high-performance liquid chromatography.

A reversed-phased HPLC method that allows the separation and simultaneous determination of the preservatives benzoic (BA) and sorbic acids (SA), methyl- (MP) and propylparabens (PP) is described. The separations were effected by using an initial mobile phase of methanol-acetate buffer (pH 4.4) (35:65) to elute BA, SA and MP and changing the mobile phase composition to methanol-acetate buffer (p...
